Methodology: Flow Cytometry
Test Description
Staining for CD45 detects most cells of hematopoietic origin at all stages of maturation. The combination of CD45 intensity and side-scatter is used in flow cytometry for acurate separation of the main leukocyte populations in bone marrow and peripheral blood samples, and for detection of abnormal cell populations. CD45 expression is regarded as a highly useful marker of hematopoietic lineage in staining panels for poorly differentiated neoplasms. However, some hematopoietic neoplasms may be negative for CD45. These include myelomas, some plasmablastic lymphomas, some acute myeloid and lymphoblastic leukemia cases, and some cases of T-anaplastic large cell lymphoma.
Specimen Requirements
Testing for this antigen is typically performed as a part of the leukemia/lymphoma screening panel. For customized testing panels including this antigen, please contact the Medical Director of the Flow Cytometry laboratory.
Storage and Transport
Maintain specimen at room temperature. Do not refrigerate or freeze. Use cold pack for transport, making sure cold pack is not in direct contact with specimen.
